transparent image hotspots.

sorry if this has been asked before, I really need to find a way to make the icons for image hotspots be transparent, I have over 40 course that I need to do this for and it's very fustrating now being about to do this. 

I have tried PNG transparent files as well but they show up white or black. 

is there a quick fix?
